I assume you're talking about derailleur type 5-speed. Most coaster hubs have long enough axles to add an extra pair of wheel nuts or some type of spacer to take up the extra space. Some newer ones may be too short, if so, it should be obvious at a glance. 99% of coaster sprockets will need to have a 1/8" chain instead of the 3/32" used on 5-speeds. Up front, the 3/32" sprocket will work, but 1/8" will work better and last longer.
 
Show us the frame, and let us know what hub you had in mind. You can almost certainly go singlespeed, but the build requirements/particulars are going to vary depending on the frame.
 
I'm using a 1/8 chain on a fixie 1/8 sprocket to an old "10 speed" 3/32 chain ring up front with no issues.
